She was born in San Francisco, California, United States to Puerto Rican mother and Irish father. Her mother died when she was just eleven. She has one brother and both of them were raised by her father alone. Her father Tony was real estate investor. She is American and is of white ethnicity.

She has been married twice. She married for the first time in her life to Gavin Newsom in December 2001. He is an American politician and he was elected mayor of San Francisco in 2003. The couple married at Saint Ignatius Catholic Church on the campus of the University of San Francisco, where Guilfoyle attended law school. After five years of marriage in January 2005, they both filed for divorce. The reason for the divorce they mentioned was bi-coastal marriage. Their divorced was finalized in 2006.

Soon after her divorced was finalized, she got married for second time to Eric Villency. He is the CEO of Villency Design Group, an interior and product design firm. They got married on the island of Barbodas on 27 May, 2006. They have one child together. The couple announced they are separated and filed for divorce on June 23, 2009. Their divorced was finalized in 2010. It is rumored that, she has numbers of love affairs after her divorced but there is no any proof about that.
